Abstract
Food, water, and environmental microbiology play a vital role in protecting public health by detecting, quantifying, and characterizing microorganisms that influence product quality, environmental safety, and the transmission of infectious diseases. Microbiological quality assessment provides essential information regarding contamination, hygiene, spoilage, and the presence of potential pathogens in food products, drinking water, wastewater, soil, air, and other environmental samples. This chapter presents the fundamental principles and practical methods used for microbiological examination across food, water, and environmental settings. Representative sampling, aseptic collection, transportation, storage, sample preparation, serial dilution, and appropriate culture techniques are discussed as critical components of reliable analysis. Conventional quantitative methods, including standard plate count, viable microbial count, membrane filtration, and the most probable number (MPN) technique, are described for assessing microbial loads and indicator organisms. Particular emphasis is placed on the detection of coliforms, Escherichia coli, enteric pathogens, foodborne microorganisms, spoilage organisms, and other indicators of sanitary quality. Microbiological examination of air, surfaces, soil, and water is also considered in relation to environmental monitoring and contamination control. Rapid and advanced approaches, including immunological assays, automated systems, polymerase chain reaction-based detection, and other molecular methods, are introduced as complementary tools for improved sensitivity and turnaround time. Quality assurance, appropriate controls, biosafety, interpretation of microbial limits, and adherence to applicable microbiological standards are emphasized throughout. Effective microbiological quality assessment contributes to food safety, water security, environmental surveillance, outbreak prevention, sanitation monitoring, and overall protection of human and community health.
