Abstract

Isolation, cultivation, and preservation of microorganisms are fundamental laboratory practices in microbiology that enable the recovery, characterization, identification, and long-term maintenance of microbial cultures. This chapter presents the essential principles and practical approaches used for obtaining pure microbial cultures from clinical, environmental, food, water, and other biological samples. Various isolation techniques, including streak plate, spread plate, pour plate, serial dilution, enrichment culture, and selective cultivation methods, are discussed with emphasis on their applications and limitations. The chapter also describes the nutritional and environmental requirements necessary for microbial growth, including appropriate culture media, temperature, pH, oxygen availability, moisture, and incubation conditions. Methods for cultivating aerobic, anaerobic, facultative, and fastidious microorganisms are addressed along with procedures for evaluating colony morphology and culture characteristics. Particular attention is given to aseptic techniques, prevention of contamination, and maintenance of pure cultures during routine laboratory procedures. Short-term and long-term preservation strategies, such as periodic subculturing, refrigeration, mineral oil overlay, freezing, cryopreservation, and lyophilization, are explained with their respective advantages and applications. The importance of culture collections, strain authentication, viability assessment, genetic stability, biosafety, and quality-control measures is also highlighted. Understanding these techniques provides students, researchers, and laboratory professionals with essential practical competencies for maintaining reliable microbial cultures and supporting diagnostic microbiology, biotechnology, research, and industrial applications.
