Abstract
Accurate preparation and standardization of laboratory reagents, buffers, and solutions are fundamental to reliable biochemical experimentation and clinical laboratory analysis. The quality of analytical results depends substantially on the correct selection of chemicals, precise measurement of solutes and solvents, appropriate concentration calculations, controlled pH, and proper storage of prepared solutions. This chapter presents the essential principles and practical procedures involved in preparing commonly used laboratory reagents, including molar, molal, normal, percentage, and dilute solutions. It explains calculations related to molecular weight, equivalents, dilution factors, concentration conversion, and preparation of working solutions from concentrated stock solutions. Particular emphasis is placed on buffer systems, their mechanism of action, buffer capacity, pKa, and the application of the Henderson–Hasselbalch equation in selecting and preparing buffers of desired pH. The principles of reagent standardization are discussed with reference to primary and secondary standards, volumetric analysis, titration procedures, standard curves, and verification of reagent performance. Factors affecting reagent stability, such as temperature, light, moisture, contamination, oxidation, and storage duration, are also considered. Appropriate labeling, documentation, preparation dates, expiry dates, storage requirements, and safe handling practices are highlighted as integral components of good laboratory practice. Quality-control measures and standardized operating procedures are emphasized to minimize preparation errors and maintain consistency between analytical procedures. A thorough understanding of these principles enables students and laboratory professionals to prepare dependable reagents and buffer systems, thereby improving analytical accuracy, reproducibility, laboratory safety, and the overall quality of biochemical investigations.
